The transcriptional co‐activators YAP (or YAP1) and TAZ (or WWTR1) are frequently activated during the growth and progression of many solid tumors, including lung, colorectal, breast, pancreatic, and liver carcinomas as well as melanoma and glioma. YAP/TAZ bind to TEAD‐family co‐activators to drive cancer cell survival, proliferation, invasive migration, and metastasis. YAP/TAZ activation may also confer resistance to chemotherapy, radiotherapy, or immunotherapy. YAP‐TEAD cooperates with the RAS‐induced AP‐1 (FOS/JUN) transcription factor to drive tumor growth and cooperates with MRTF‐SRF to promote (...) activation of cancer‐associated fibroblasts, matrix stiffening, and metastasis. The key upstream repressor of YAP/TAZ activation is the Hippo (MST1/2‐LATS1/2) pathway and the key upstream activators are mechanically induced Integrin‐SRC and E‐cadherin‐AJUBA/TRIP6/LIMD1, growth factor induced PI3K‐AKT, and inflammation‐induced G‐protein coupled receptor (GPCR) signals, all of which antagonize the Hippo pathway. In this review, strategies to target YAP/TAZ activity in cancer are discussed along with the prospects for synergy with established pillars of cancer therapy. (shrink)

Several lines of evidence have underscored the remarkable neuroplasticity of the primate sensorimotor cortex, characterizing these cortical areas as dynamic constructs that are modelled in a use-dependent manner by behaviourally significant experiences. Their plasticity likely provides a neural substrate that may contribute to the dynamic systems paradigm argued by Shanker & King (S&K) as crucial for development of communication skills.

This commentary focusses on MacNeilage's arguments and evidence that the development of cerebral cortical controls over cyclic ingestive movements has provided substrates for the evolution of speech production. It outlines evidence from experimental approaches using cortical stimulation, inactivation, and single neuron recording in primates that lateral frontal cortical regions are indeed crucial for the generation and guidance of cyclic orofacial movements.

Traditional combinatory logic uses combinators S and K to represent all Turing-computable functions on natural numbers, but there are Turing-computable functions on the combinators themselves that cannot be so represented, because they access internal structure in ways that S and K cannot. Much of this expressive power is captured by adding a factorisation combinator F. The resulting SF-calculus is structure complete, in that it supports all pattern-matching functions whose patterns are in normal form, including a function that decides structural equality (...) of arbitrary normal forms. A general characterisation of the structure complete, confluent combinatory calculi is given along with some examples. These are able to represent all their Turing-computable functions whose domain is limited to normal forms. Organizational ethics programs often are created to address tensions in organizational values that have been identified through repeated clinical ethics consultation requests. Clinical ethicists possess some core competencies that are suitable for the leadership of high-quality organizational ethics programs, but they may need to develop new skills to build these programs, such as familiarity with healthcare delivery science, healthcare financing, and quality improvement methodology. To this end, we suggest that clinical ethicists build organizational ethics programs incrementally and via quality improvement (...) projects undertaken in collaboration with senior clinical leaders. Organizational ethics programs often differ from clinical ethics programs in their membership and processes, and likely will require ethicists to forge new partnerships with a wide array of organizational leaders. With attention to the ways that organizational ethics programs differ from clinical ethics programs, and investment in quality improvement methodology and formal institutional needs assessments, clinical ethics leaders can position an organizational ethics program to advocate effectively for visible and compelling alignment of leadership decision making with the values of the organization. Each paper is printed in full and is followed by a critical discussion which raises questions of statistical treatment, sufficiency of controls and alternative interpretations. Understanding spoken words involves a rapid mapping from speech to conceptual representations. One distributed feature-based conceptual account assumes that the statistical characteristics of concepts’ features—the number of concepts they occur in and likelihood of co-occurrence —determine conceptual activation. To test these claims, we investigated the role of distinctiveness/sharedness and correlational strength in speech-to-meaning mapping, using a lexical decision task and computational simulations. Responses were faster for concepts with higher sharedness, suggesting that shared features are facilitatory in tasks like lexical decision (...) that require access to them. Correlational strength facilitated responses for slower participants, suggesting a time-sensitive co-occurrence-driven settling mechanism. The computational simulation showed similar effects, with early effects of shared features and later effects of correlational strength. These results support a general-to-specific account of conceptual processing, whereby early activation of shared features is followed by the gradual emergence of a specific target representation. This article discusses the meaning of ecclesiology and church polity as such, in relation to each other, but also in connection to church order which lies in the elongation of both. From a reformed perspective, it is indicated that the ecclesiology of the Netherdutch Reformed Church over the last fifty to hundred years, centred round the viewpoint of the Church as a volkskerk, in conjunction with the church orderly, of the Kerkwet till 1997. Ephesians 1:14 and 1:22, 23 as a scriptural maximum-minimum for ecclesiology. Against the background of a membership decline in mainstream churches in South Africa and abroad, and conscious of the theological debate in which a missional ecclesiology is presented as at least part of the modern-day solution to the problem, this article points in the direction of a Christological ecclesiology as an important ecclesiological point of departure. The article gives an exegetical outline of Ephesians 1:14 and 1:22, 23 as a (...) critical maximum-minimum for a Christological ecclesiology and as a means of making missional ecclesiology more acceptable. A tolerant treatment of church order and the law of usance: This article discusses the recent decline of church polity in the Netherdutch Reformed Church which is obvious in different areas of the Church with special reference to the liturgy practised in sermons of some congregations. This decline is also observed in other churches in South Africa and abroad. The article is a reaction to the reason for this situation and indicates that it should not be found in a collegialistic (...) concept of church, but rather due to congregationalism or independentism in modern form. The article’s viewpoint is that the above-mentioned church polity decline is a result of the fact that church order is not valued as an order with a Scriptural and therefore an ecclesiological basis. Despite its importance, the linkage between genetic and cultural evolution has until now been little explored. An understanding of this linkage is needed to extend evolutionary theory so that it can deal for the first time with the phenomena of mind and human social history. We characterize the process of gene-culture coevolution, in which culture is shaped by biological imperatives while biological traits are simultaneously altered by genetic evolution in response to cultural history. A case is made from both theory (...) and evidence that genetic and cultural evolution are inseverable, and that the human mind has tended to evolve so as to bias individuals toward certain patterns of cognition and choice rather than others. With the aid of mathematical models we trace the coevolutionary circuit: The genes prescribe structure in developmental pathways that lay down endocrine and neural systems, imposing regularities in the development of cognition and behavior; these regularities translate upward into holistic patterns of culture, which can be predicted in the form of probability density distributions ; natural selection acts within human history to favor certain epigenetic rules over others; and the selection alters the frequencies of the underlying genes. The effects of genetic and cultural changes reverberate throughout the circuit and are consequently tested with the passage of each life cycle. In addition to modeling gene-culture coevolution, we apply methods from island biogeography and information theory to examine the cultural capacity of the genes, the factors determining the magnitude of cultural diversity, and the possible reasons for the uniqueness of the human achievement. The view that it is better for life to be created free of disability is pervasive in both common sense and philosophy. We cast doubt on this view by focusing on an influential line of thinking that manifests it. That thinking begins with a widely-discussed principle, Procreative Beneficence, and draws conclusions about parental choice and disability. After reconstructing two versions of this argument, we critique the first by exploring the relationship between different understandings of well-being and disability, and the second (...) by more briefly focusing on the idea of a significant reason. By placing these results against the broader historical and ongoing contexts in which the lives of those with disabilities have been deemed of inferior quality, we conclude with a call for greater humility about disability and well-being in thought and practice. This article gives a short reflection on the background and origin of the social-scientific model change agent in relation to change agency, a relationship that causes an alternate state of consciousness for the change agent. Arguing from the seven-point explanation of Malina and Pilch of how a change agent operates, this article applies the model to the letter to the Ephesians. The conclusion is that this social-scientific model is helpful for the recent church to realise how narrow the relation is (...) between Jesus Christ and the church, in view of the future of the church as a future in Christ. A choice is made to find alternate words in the Afrikaans language suitable for the theological debate, namely opdraggewer [change agency], meningsvormer [change agent] and alternatiewe staat van bewussyn [alternate state of consciousness].
